Ready-to-use pNPP Substrates
Kem-En-Tec Diagnostics pNPP (p-NitroPhenyl Phosphate) substrates are liquid buffer solutions containing stabilized pNPP used for measuring Alkaline Phosphatase activity. Current methods for making pNPP-solutions involve dissolving the pNPP powder in proper buffers or diluting pNPP concentrates. The stability of these solutions is normally one day. By contrast, our ready-to-use mono-component substrates are stable up to 36 months from manufacture date when stored at 2-8°C, without loss of activity (for details see below).

No harmful components
Kem-En-Tec Diagnostics pNPP substrates are aqueous based and contain NO harmful organic solvent, including 1-methyl-2-pyrrolidone (NMP) which was added to the REACH Restricted Substances List in May 2018. The non-hazardous stop solution recommended for stopping the pNPP reaction does not require harmful labeling – enabling worldwide shipping of the RTU ELISA kits.
In the presence of AP, pNPP is hydrolyzed rapidly to p-nitrophenol and inorganic phosphate. The reaction results in a water-soluble yellow end product (p-nitrophenol) that absorbs light at 405 nm. The reaction can be stopped by adding an equal amount of 0,1 M sodium hydroxide (NaOH) solution and the absorbance can be read at 405 nm. The intensity of the color is a measure of the concentration of the reactant.
